چکیده
The East Indian sandalwood tree, Santalum album, yields one of the costliest heartwoods and precious essential oil. Unsurprisingly, this endangered forest species is severely affected due to unmet global demands, illegal trade and harvesting, overharvesting and an epidemic mycoplasmal spike disease. In vitro micropropagation endeavours have resulted in defined in vitro stages such as somatic embryos that are amenable to mass production in bioreactors. We report on somatic embryo production in a 10-L air-lift-type bioreactor, and compare the growth and biochemical parameters with those of a 2-L air-lift-type bioreactor. For the 10-L bioreactor with biomass (475.7+18 g fresh weight; P , 0.01), concomitantly santalols (5.2+0.15 mg L; P , 0.05), phenolics (31+1.6 mg L) and arabinogalactan proteins (AGPs) (39+3.1 mg L; P , 0.05) are produced in 28 days. In addition, we identified and quantified several santalols and phenolics by means of high-performance thin-layer chromatography and reverse-phase high-pressure liquid chromatography analyses, respectively. Results indicate that 10-L-capacity air-lift bioreactors are capable of supporting somatic embryo cultures, while the extracellular medium provides opportunities for production of industrial raw materials suchassantalols, phenolicsand AGPs.This will proveuseful for furtheroptimizationand scale-upstudiesofplant-produced metabolites.

Preparative separation of α- and β-santalenes and (Z)-α- and (Z)-β-santalols using silver nitrate-impregnated silica gel medium pressure liquid chromatography and analysis of sandalwood oil.
The major sesquiterpene constituents of East-Indian sandalwood oil (Z)-α- and (Z)-β-santalols have shown to be responsible for most of the biological activities and organoleptic properties of sandalwood oil. The work reported here describes the strategic use of medium pressure liquid chromatography (MPLC) for the separation of both α- and β-santalenes and (Z)-α- and (Z)-β-santalols. Silver nitr...

Polygalacturonase production under different shear stress environments by Aspergillus sp. was studied. It was found that the rate of enzyme production in the stirred tank fermentor decreased with increasing the stirrer speed in the range 300-750 rpm. On the other hand, in a split cylinder air-lift fermentor, the rate of polygalacturonase production slightly increased with increasing in air ...

Extended Abstract Introduction and Objective: In order to overcome the contamination problems, as well as the usual method of increasing potato through micro tuber, micropropagation has been considered through tissue culture. The aim of this study was to evaluate the callus induction and embryogenesis in potato cultivars using leaf explants. Material and Methods: For this purpose, micropropa...
متن کاملComparative stepwise pattern of reactive oxygen species production during in vitro development of fertilized and nuclear transferred goat embryos
Objective A unique feature of embryo metabolism is production of reactive oxygen species (ROS). It is well established that during in vitro culture, ROS levels increase over normal ranges observed for embryos developed in vivo. This study evaluates and compares the stepwise pattern of ROS production during in vitro development of reconstructed goat embryos produced by zona-free method of somati...
متن کاملN-acetylglucosamine and glucosamine-containing arabinogalactan proteins control somatic embryogenesis.
In plants, complete embryos can develop not only from the zygote, but also from somatic cells in tissue culture. How somatic cells undergo the change in fate to become embryogenic is largely unknown. Proteins, secreted into the culture medium such as endochitinases and arabinogalactan proteins (AGPs) are required for somatic embryogenesis. Here we show that carrot (Daucus carota) AGPs can conta...
